CT-179 is a highly potent and selective small molecule inhibitor of Olig2 inhibitor. In preclinical studies, CT-179 disrupted OLIG2 transcriptional activity, leading to impaired glioma stem-like cell proliferation and self-renewal. For example, in OLIG2+ patient-derived DIPG cell lines, CT-179 reduced cell viability with IC₅₀ values in the low micromolar range (e.g., ~1–3 μM). In mouse orthotopic xenograft models of DIPG, systemic administration of CT-179 resulted in significant tumor growth inhibition and increased median survival. Additionally, RNA-seq analyses post-treatment showed downregulation of OLIG2-regulated gene networks involved in neural progenitor maintenance and proliferation.
