Ganoderic acid C1 is a triterpenoid that has been found in G. lucidum and has diverse biological activities. It inhibits angiotensin-converting enzyme. Ganoderic acid C1 also inhibits HIV-1 protease with an IC50 value of 180 µM. It reduces the proliferation of murine Lewis lung carcinoma cells but not murine Meth-A sarcoma cells. Ganoderic acid C1 (20 µg/ml) inhibits LPS-induced NF-κB phosphorylation and production of TNF-α in RAW 264.7 cells, as well as in peripheral blood mononuclear cells (PBMCs) isolated from patients with asthma.

Biological target:
Ganoderic acid C1, a natural compound that could be isolated from G. lucidum, suppresses TNF-α production by murine macrophages (RAW 264.7 cells).
In vitro activity:
This study first established the inhibitory effects of GAC1 (ganoderic acid C1) on TNF-α production by LPS stimulated-RAW 264.7 macrophages. LPS (1μg/mL) enhanced TNF-α production, and pretreatment with GAC1 decreased this TNF-α production in a dosedependent manner with a maximal suppressive effect at 20 and 40μg/mL. In vivo activity:
