Biological target:
Dioscin(CCRIS 4123; Collettiside III) is a natural steroid saponin derived from several plants, showing potent anti-cancer effect against a variety of tumor cell lines.
In vitro activity:
Dioscin also alleviated the release of inflammatory cytokines in response to LPS in microglial BV-2 cells (Figure S3). Further, we used the DAPI staining technique to validate dioscin-mediated protection against MPP+-induced apoptosis on SH-SY5Y (Figure 2B). MPP+ treatment showed significant chromatin fragmentation within the nucleus without changing nuclear phenotype. Pre-treatment of dioscin 200 and 400 ng/mL, but not 100 ng/mL, dose significantly rescued from apoptotic cell death. This result indicates that dioscin improves neuronal cells’ survivability by reducing MPP+-induced mitochondrial complex I impairment. In vivo activity:
DIO (dioscin) increased the stool number, stool water content and shortened whole gut transit time, helped to recover the gut microbial diversity and microbiota community structure, and increased the abundance of Muribaculaceae in STC mice. These results provide that DIO increases GI motility and inhibits neuron loss by modulating gut microbiota profile, improving inflammation in the ENS microenvironment and up-regulating the BMP2 secreted by MMs. Reference: Iran J Basic Med Sci. 2022 Sep;25(9):1132-1140. https://pubmed.ncbi.nlm.nih.gov/36246057/
