JXL069 is a potent and selective mitochondrial pyruvate carrier (MPC) inhibitor that disrupts pyruvate import into mitochondria, thereby modulating cellular metabolism. In biochemical assays, JXL069 demonstrated submicromolar inhibitory activity against MPC, with an IC₅₀ value of approximately 0.53 µM in MPC1/2-mediated pyruvate transport assays. Cellular studies showed that JXL069 effectively reduces mitochondrial oxygen consumption and pyruvate-driven respiration in cancer cell lines, with an EC₅₀ around 1–2 µM depending on the cell type. Functionally, treatment with JXL069 induces a metabolic shift toward glycolysis and impairs proliferation in MPC-dependent tumor cells, supporting its potential as a chemical probe for studying mitochondrial metabolism and as a lead compound for targeting metabolic vulnerabilities in cancer.
