HY50737A is a selective, non-covalent inhibitor of the deubiquitinase USP8, binding to its catalytic domain to block substrate access and disrupt protein homeostasis. It demonstrates potent bioactivity with an IC₅₀ of 0.28 µM in enzymatic assays targeting USP8. In MCF7 breast cancer cells, treatment with 0.1 µM HY50737A for 24 hours reduces proliferative capacity and induces transcriptomic changes consistent with impaired deubiquitination.
