KKL-35 is a small-molecule inhibitor targeting the bacterial trans-translation pathway, an essential ribosome rescue system absent in mammals, making it a promising antibacterial candidate. KKL-35 has been shown to inhibit Escherichia coli trans-translation with a reported MIC (minimum inhibitory concentration) range of 0.5–2 µg/mL against various E. coli clinical isolates. It also displays activity against other Gram-negative and Gram-positive bacteria, including Pseudomonas aeruginosa (MIC ~8–16 µg/mL) and Staphylococcus aureus (MIC ~0.25–2 µg/mL). Mechanistically, KKL-35 blocks the binding of tmRNA-SmpB to stalled ribosomes, impairing bacterial viability.
