Biological target:
Liraglutide is a glucagon-like peptide-1 (GLP-1) receptor agonist used clinically to treat type 2 diabetes mellitus.
In vitro activity:
To determine the effect of liraglutide on oxLDL-induced foam cell formation, Raw 264.7 cells were exposed to oxLDL (50 µg/mL) in the presence, or absence of liraglutide (1 nmol/L) for 48 h, Oil Red O staining was then performed. In vivo activity:
The present study explored the effects of an anti-diabetic drug, liraglutide (LIR), in reversing major pathological hallmarks in the prodromal disease stage of both the 5xFAD transgenic and SAD (sporadic AD mouse models of AD (Alzheimer’s Disease). The results indicated that STZ (streptozotocin) significantly increased neuroinflammation, Aβ plaque deposition and disrupted insulin signaling pathway, while 25 nmol/kg LIR, when injected IP, significantly decreased neuroinflammatory responses in both SAD and 5xFAD mice before significant cognitive changes were observed, suggesting LIR can reduce early neuropathology markers prior to the emergence of overt memory deficits.
