Biological target:
Stattic selectively inhibits STAT3 activation, dimerization, and nuclear translocation by preventing the binding of tyrosine-phosphorylated peptide motifs to the STAT3 SH2 domain (IC50 = 5.1 μM at 37°C).
In vitro activity:
This study proposes Stattic as a tool for the inhibition of STAT3 in cell lines or animal tumor models displaying constitutive STAT3 activation. Stattic selectively inhibited activation, dimerization, and nuclear translocation of STAT3 and increases the apoptotic rate of STAT3-dependent breast cancer cell lines. In vivo activity:
Stattic could be a promising target for management of cardiac sepsis and inflammation-related cardiac damage. In a murine polymicrobial sepsis model, Stattic markedly reversed the CLP-induced cardiac abnormalities and restored the cardiac function by its anti-inflammatory activities.
