Biological target:
Sonidegib metabolite M48 is the main circulating metabolite of Sonidegib. Sonidegib is a hedgehog pathway inhibitor. M48 showed a much longer Tmax (60 h) than Sonidegib.
In vitro activity:
Sonidegib significantly suppressed proliferation in NKTCL cells and the effect was dose-dependent. Further analysis revealed that sonidegib treatment elevated the number of apoptotic cells in a dose- and time-dependent manner. In vivo activity:
Consistent with these findings, orally administered sonidegib attenuated IL-6 and TNF-α levels in the brains of LPS-treated mice. Collectively, these results indicate that sonidegib suppresses inflammation and cell migration in LPS-treated BV2 cells and mice by inhibiting JNK and NF-κB signaling.
