PS-48 is a phosphoinositide-dependent protein kinase-1 (PDK1) activator. Treatment of low cloning efficiency BFFs with PS48 resulted in an increase in the intracellular lactate production and H3K9 acetylation, decrease in histone deacetylase activity and HP1α expression, less condensed chromatin structure and more cloning embryos developing to blastocysts.

Biological target:
PS-48 is a PDK1 activator and binds to the HM/PIF binding pocket rather than the ATP-binding site.
In vitro activity:
The use of 10 µM PS-48 significantly increased the proportion of porcine oocytes reaching the metaphase II (MII) stage and improved cumulus cell expansion. The activation of the PI3K/Akt pathway was associated with the regulation of maturation-related genes and proteins, including Akt, cyclin B1, MOS, BMP15, GDF9, CDC2, mTOR, BAX, BCL2, and caspase-3. In vivo activity:
In a mouse model for Alzheimer's disease, the oral administration of PS-48 was tested to assess its potential in preventing the decline in learning and memory. PS-48 had a positive impact on spatial learning and memory, especially in the mice on a high-fat die. PS-48 modestly reduced body weight and improved glucose tolerance.
