Biological target:
Roslin-2 impairs the interaction between focal adhesion kinase (FAK) and p53 and restores p53 transcriptional activity. It reverses FAK-mediated suppression of the expression of p53-dependent p21, MDM2, and Bax in wild-type p53 HCT116 colon and MCF-7 breast cancer cells. Roslin-2 reduces tumor growth and activates p21 in p53+/+, in HCT116 mouse xenograft models.
In vitro activity:
Roslin-2 increased p53 transcriptional activity that was inhibited by FAK using p21, Mdm-2, and Bax-promoter targets. Roslin-2 also caused increased expression of p53 targets: p21, Mdm-2 and Bax proteins. Reference: BMC Cancer. 2013 Jul 11;13:342. https://pubmed.ncbi.nlm.nih.gov/23841915/
In vivo activity:
Roslin-2 significantly decreased tumor growth, disrupted the complex of FAK and p53, and up-regulated p21 in HCT116 p53+/+ but not in HCT116 p53-/- xenografts in vivo.
