ARDP0006 is an anthraquinone‐based inhibitor targeting the dengue virus NS2B–NS3 serine protease. In vitro, it demonstrated the strongest enzymatic inhibition among 15 tested compounds, reducing dengue viral titer by over 1 log PFU/mL in cell‑based assays. Structural studies indicate ARDP0006 binds to the protease’s allosteric site, disrupting NS2B–NS3 activity more effectively than its parent compound. In addition to potent enzymatic inhibition, it showed antiviral efficacy in cellular systems, making it a lead scaffold for flavivirus protease inhibitors.
