Biological target:
SNAP-94847 is a potent melanin-concentrating hormone receptor 1 (MCH1) antagonist (Ki = 2.2 nM, KD = 530 pM) that displays > 80-fold and > 500-fold selectivity over α1A and D2 receptors respectively. SNAP-94847 increases neurogenesis in the dentate gyrus and decreases food-reinforced operant responding in vivo. SNAP-94847 exhibits anxiolytic activity.

These findings suggest that the combination of rimonabant and SNAP-94847 is highly effective in reducing the body weight and normalizing behavioral effects of obese mice. In lean mice, both drugs independently reduced food intake. When administered together to obese mice, the combination resulted in a more significant weight loss than individual treatments. The combination therapy also influenced markers of metabolism in adipose tissues, indicating increased energy expenditure and fat breakdown.
