Biological target:
SC75741 is an NF-κB inhibitor with an IC50 of 200 nM for p65, resulting in reduced expression of cytokines, chemokines, and pro-apoptotic factors. SC75741 blocks the replication of influenza viruses (IV). SC75741 inhibits caspase activation and blocks caspase-mediated nuclear export of viral ribonucleoproteins.
In vitro activity:
SC75741 has potential for targeting the HBx viral protein (associated with the hepatitis B virus) and potentially combating HBV-associated liver conditions. Of 179 compounds studied, SC75741 had a very low docking energy, making it one of the most promising compounds. During simulations, SC75741 robustly inhibited HBx. It exhibited stable interaction and appeared to block a critical region (aa 88-100) near the HBx helix linked to cell invasion. In vivo activity:
SC75741 has beneficial neuroprotective effects in amyotrophic lateral sclerosis (ALS). SC75741 inhibited NF-κB, which inhibited inflammation-induced aggregation. SC75741 degraded pre- aggregated proteins by inhibiting the c-Abl mediated autophagy-lysosomal pathway. SC75741 enhanced the interaction between p62 with TDP25 and LC3C, promoting TDP25 degradation.
