Xanthohumol shows anti mutagenic activity against mutations induced by the food borne mutagen 2-amino-3-methylimidazo[4,5-f]quinoline (IQ). The mechanism is possibly related to the inhibition of the metabolic activation of IQ by human cytochrome P450 1A2 (CYP1A2) and the binding of IQ metabolites to DNA and proteins.

In vitro activity:
Thymol treatment reduced the viability of F. nucleatum and P. gingivalis. The presence of these vegetable extracts during the development of a dynamic in vitro multispecies biofilm model inhibited the filamentous growth of C. albicans, partially reversing the effect that the fungus exerted on the structure, size and vitality of periodontopathogenic bacteria. In vivo activity:
XN significantly reduced adenocarcinoma formation and inflammation in vivo while decreasing IKKβ and NF-κB signaling in both models. XN binds to the C179 residue of IKKβ, inhibiting its activity. Additionally, our findings highlight KEAP1's role as an upstream regulator of IKKβ degradation. XN specifically interacts with the C288 residue of KEAP1, showing triple-binding affinity with IKKβ and KEAP1.
