PNU-101017, also known as U-101017, is a gamma-aminobutyric acid (GABA) receptor agonist potentially for the treatment of anxiety. PNU-101017 has a high affinity for the GABAA receptor subtypes containing the alpha 1 and alpha 3 or alpha 5 subunits. PNU-101017 was weakly active in anti-conflict anxiolytic tests, but attenuated the plasma corticosteroid response to mild stress in rats. PNU-101017 did not potentiate the CNS-depressant effects of ethanol, and produced no evidence of physical dependence when administered repeatedly.
