Biological target:
11-Beta-hydroxyandrostenedione is a 11β-hydroxysteroid dehydrogenase (11βHSD) isozymes inhibitor.
In vitro activity:
5–8F and NP69 cell lines were treated with 25, 50, or 100 μg/mL NPC01 for 24 or 48 h and measured using Annexin V–FITC/PI staining to investigate the possible apoptotic effect of NPC01 on NPC cells. Compared with the control group, NPC cell lines treated with NPC01 showed a significant increase in cell apoptotic population in a dose-dependent manner and 25 μg/mL which has no statistical significance when compared to the control (Data not shown) (Figure 2). In vivo activity:
In ovariectomized rats, 11betaOHA (11beta hydroxy-androstenedione) restored osteogenesis and increased the bone mineral density at both the metaphyseal and diaphyseal regions of the femur. Bone histomorphometric study of ovariectomized rats demonstrated that the mineral apposition rate of both cortical bone and trabecular bone was increased by treatment with 11betaOHA. In addition, 11betaOHA increased alkaline phosphatase activity in cultured osteoblastic cells (MC3T3-E1 and SaOS-2).
