AZD6538 is a negative allosteric modulator (NAM) of metabotropic glutamate receptor 5 (mGluR5), a G-protein-coupled receptor (GPCR) predominantly expressed in the central nervous system. mGluR5 plays a key role in modulating synaptic plasticity, learning, memory, and excitatory neurotransmission. NAMs like AZD6538 bind to a site distinct from the receptor’s orthosteric ligand-binding site, reducing receptor activity without completely blocking its function
